The present invention relates to a pharmaceutical composition comprising a TIS21 expression promoter or activator as an active ingredient for preventing or treating mDia (diaphanous-related formin)-related diseases and a method for screening the same. According to the present invention, it is determined that an increase in activity of Akt1 and a decrease in expression of Sp1 are induced in breast cancer cells in which TIS21 is over-expressed, and a decrease in activated Akt1 and expression of Sp1 inhibits mDia family genes. Thus, the composition comprising a TIS21 expression promoter or activator as an active ingredient can be used as a pharmaceutical composition for preventing or treating mDia gene-related diseases. When, using a method for screening a TIS21 expression stimulator or activator in a tissue of a patient who shows an increase in expression of mDia, it is possible to screen a drug stimulating or activating expression of TIS21 specifically. Therefore, it is possible to develop an agent for treating mDia gene-related diseases effectively.COPYRIGHT KIPO 2016본 발명은 TIS21 발현 촉진제 또는 활성화제를 유효성분으로 함유하는 mDia (Diaphanous-related formin) 관련 질환 예방 또는 치료용 약학조성물 및 이의 스크리닝 방법에 관한 것으로, 본 발명에 따르면 TIS21가 과발현된 유방암 세포에서는 Akt1의 활성 증가 및 Sp1의 발현 감소가 유도되었으며, 활성화된 Akt1 및 Sp1 발현 감소는 mDia 패밀리 유전자의 발현을 억제시키는 것을 확인함에 따라, 본 발명의 TIS21 발현 촉진제 또는 활성화제를 유효성분으로 함유하는 조성물은 mDia 유전자관련 질환 예방 또는 치료용 약학조성물로 사용될 수 있으며, mDia 발현이 증가된 환자의 조직에서 TIS21 발현 촉진제 또는 활성화제의 스크리닝 방법을 이용하면 TIS21 발현을 촉진 또는 활성화하는 약물을 특이적으로 선별할 수 있어, mDia 유전자 관련 질환 치료제를 효과적으로 개발할 수 있다.
