PROBLEM TO BE SOLVED: To remove platelets from apheresis blood.SOLUTION: Monocytes are prepared so as not to include platelets. The present invention provides a method of preparing monocytes including a step of: (1) preparing apheresis blood to which is blended a non-cytotoxic, non-ionic density regulator such that the density of the blood plasma is 1.066 g/ml to 1.078 g/ml； and (2) separating monocytes by centrifuging a layered product comprising the following layer a and layer b. Layer a: density gradient centrifugation medium for separating monocytes.
