The processing cells, methods, systems and kits are provided for example therapeutic uses, such as adoptive cell therapy according to the present invention. The method provided comprises a transduction method of conditions which results in transduction of the cells with the viral vector, incubating the cells and viruses. In some embodiments the incubation is carried out in the cavity of the substantially rigid distal chambers, such as a cylindrical chamber made of hard plastic, the cavity may have a variable volume. The method of the present invention includes other processing steps that are carried out within such a chamber, such as washing, screening, isolation, culture and formulated. In particular, the invention relates to a method of providing a number of advantages over conventional processing methods such as the methods available in large-scale processing.
