The present invention relates to a composition containing a herbal medicine extract mixture as an active ingredient for the prevention, alleviation, or treatment of arthritis, and more specifically, to a composition containing a herbal medicine extract mixture as an active ingredient for the prevention, alleviation, or treatment of arthritis, which can be used as heath functional food or a pharmaceutical composition. According to the present invention, the herbal medicine extract mixture (YNH431) inhibits the expression of ERK, p38 MAPK, and NF-kB caused by the excessive proliferation of synovial fibroblasts, thereby having an inflammation inhibitory effect and an excellent inhibitory effect against the differentiation of osteoclasts associated with a fracture in a joint. It is verified that the expression rate of MMPs, COX-2, and PGE2 increased by inflammation is further reduced in the case of using the herbal medicine extract mixture of the present invention than in the case of separately using each herbal medicine extract. Furthermore, the expression of IL-1&beta and TNF-&alpha, which are inflammatory cytokines, is inhibited in animal models of inflammatory arthritis, thereby inhibiting the generation of arthritis.
