Disclosed herein is the development of an attenuated microbial vector for improved delivery of multiple targeted antigens via the insertion of cloned glutamate-dependent acid resistance genes into the attenuated microbial vector strain Salmonella enterica serovar Typhi strain Ty21a. The resulting Ty21a strain expresses the Gad A, B, and C proteins under an arabinose inducible promoter. In the presence of arabinose and under other acid tolerance-inducing conditions, such as growth at about pH 5.5, the Ty21a(Gad) strain tolerates exposure to pH 2.5 for 3 hrs. This acid resistance expression is sufficient for formulation of the vaccine as a wafer or tablet for human administration. Such formulation may occur without the need for buffer addition. Furthermore, such formulation enhances the ability of the immunogenic composition and/or vaccine to stimulate immune responses sublingually and throughout the intestinal tract and the ability of young children to easily swallow the composition and/or vaccine.La présente invention porte sur le développement dun vecteur microbien atténué pour une administration améliorée de multiples antigènes ciblés par lintermédiaire de linsertion de gènes de résistance aux acides glutamate-dépendants clonés, dans la souche de vecteur microbien atténué Salmonella enterica sérotype Typhi souche Ty21a. La souche Ty21a ainsi obtenue exprime les protéines Gad A, B et C sous un promoteur inductible par larabinose. En présence darabinose et dans dautres conditions induisant une tolérance aux acides, telles quune croissance à environ pH 5,5, la souche Ty21a(Gad) tolère lexposition à pH 2,5 pendant 3 h. Cette expression de résistance aux acides est suffisante pour la formulation du vaccin sous forme dun cachet ou comprimé destiné à ladministration à lhomme.
