The present disclosure provides methods for preparing β-substituted tryptophan compounds. The methods include: combining i) an unsubstituted indole or a substituted indole, ii) a β-substituted serine, and iii) a tryptophan synthase β-subunit (i.e., a TrpB)； and maintaining the resulting mixture under conditions sufficient to form the β-substituted tryptophan. The TrpB contains at least one amino acid mutation which promotes formation of an amino-acrylate intermediate. New TrpB variants and new β-substituted tryptophan analogs are also described.La présente divulgation porte sur des procédés de préparation de composés de tryptophane β-substitués. Les procédés comprennent : la combinaison de i) un indole non substitué ou un indole substitué, ii) une sérine β-substituée, et iii) un sous-motif β de tryptophane synthase (à savoir, TrpB)； et le maintien du mélange obtenu dans des conditions suffisantes pour former le tryptophane β-substitué. Le TrpB contient au moins une mutation d'acide aminé qui favorise la formation d'un intermédiaire amino-acrylate. De nouveaux variants de TrpB et de nouveaux analogues de tryptophane β-substitués sont en outre décrits.
