The present invention relates to a stable and readily usable form of injection filled with a mesenchymal stem cell composition cultured inside a hydrogel or, more particularly, a mesenchymal stem cell composition originated from human fat, and to a preparation method of the same. More particularly, in the present invention, provided is a composition in a readily injectable form by culturing the mesenchymal stem cells in a hydrogel, rinsing the mesenchymal stem cells, and filling a syringe with the rinsed mesenchymal stem cells, thereby eliminating a necessity of enzymatic treatment in the process of preparing final cells to be transplanted.
