PROBLEM TO BE SOLVED: To provide a practical melanoma metastasis inhibitor or a melanoma metastasis inhibition method to provide practical use for preparation of the melanoma metastasis inhibitor and to provide a practical screening method of the melanoma metastasis inhibitor.SOLUTION: Concerning the melanoma metastasis inhibitor, the melanoma metastasis inhibition method, and the use for preparation of the melanoma metastasis inhibitor, a SIRTI inhibitory substance is used. The screening method for the melanoma metastasis inhibitor includes processes for: measuring histone deacetylase activity of SIRTI in the presence of a substance comparing the value of histone deacetylase activity acquired as the result of measurement with the value of the histone deacetylase activity of SIRTI in the absence of the substance and evaluating the substance as a melanoma metastasis inhibitor, when the value of the histone deacetylase activity of SIRTI in the presence of the substance is lower than the value of the histone deacetylase activity of SIRTI in the absence of the substance.COPYRIGHT: (C)2012,JPO&INPIT【課題】本発明は、実用的なメラノーマ転移抑制剤やメラノーマ転移抑制方法を提供することや、実用的なメラノーマ転移抑制剤調製のための使用を提供することや、メラノーマ転移抑制剤の実用的なスクリーニング方法を提供することを目的とする。【解決手段】メラノーマ転移抑制剤やメラノーマ転移抑制方法やメラノーマ転移抑制剤調製のための使用については、SIRT1阻害作用物質を用いる。また、メラノーマ転移抑制剤のスクリーニング方法については、被検物質の存在下で、SIRT1のヒストンデアセチラーゼ活性を測定する工程;測定の結果得られたヒストンデアセチラーゼ活性の値を、被検物質の非存在下におけるSIRT1のヒストンデアセチラーゼ活性の値と比較する工程;及び、被検物質の存在下におけるSIRT1のヒストンデアセチラーゼ活性の値が、被検物質の非存在下におけるSIRT1のヒストンデアセチラーゼ活性の値より低い場合に、その被検物質をメラノーマ転移抑制剤と評価する工程;を用いる。【選択図】なし
