The present invention relates to Enzyme Replacement Therapy (ERT) based on phenylalanine hydroxylase (PAH) and compositions intended for this use. It concerns an erythrocyte encapsulating PAH, especially in suspension in a pharmaceutically acceptable carrier or vehicle, a pharmaceutical composition comprising erythrocytes encapsulating PAH in a pharmaceutically acceptable carrier or vehicle, and such a pharmaceutical composition for use in the treatment or prevention of phenylketonuria (PKU) and/or other diseases involving a too high level of phenylalanine； the treatment or prevention may be in combination with a Phe-restricted diet. The invention particularly relates to classic PKU, variant PKU and non-PKU hyperphenylalaninemia.La présente invention concerne une enzymothérapie substitutive (ERT) basée sur de la phénylalanine hydroxylase (PAH) et des compositions conçues pour son utilisation. L'invention concerne un érythrocyte encapsulant de la PAH, en particulier en suspension dans un support ou véhicule pharmaceutiquement acceptable, une composition pharmaceutique comprenant des érythrocytes encapsulant de la PAH dans un support ou véhicule pharmaceutiquement acceptable, et une telle composition pharmaceutique destinée à être utilisée dans le traitement ou la prévention de phénylcétonurie (PCU) et/ou d'autres maladies impliquant un taux trop élevé de phénylalanine ； le traitement ou la prévention peut être combiné à un régime limité en Phe.
