The present invention relates to a Transactive response(TAR) DNA binding protein-43(TDP-43)-GAL4 drosophila mutant, to a TDP-43-GAL4 and UAS-Green fluorescent protein(GFP) drosophila double mutant, and to a method for producing the same. More specifically, the present invention relates to a method for producing a TDP-43-GAL4 and Green fluorescent protein (GFP) double mutant drosophila model capable of expressing fluorescent protein in tissue for expressing TDP-43 genes and capable of confirming the expression position and aspect of drosophila TDP-43 genes by using the same. According to the present invention, a GAL4 system which is an expression system capable of overexpressing gene transcription in specific tissue is used, so a drosophila double mutant simultaneously including a fluorescent protein overexpression mutant and a tissue-specific expression mutant of a TDP-43 gene produced by separating a TDP-43 gene promoter region from a drosophila genome and a method for producing the same can be provided. The drosophila double mutant can be used as a very useful animal model for screening TDP-43-related factors and studying the functions of TDP-42 genes of drosophila and the functions of human degenerative brain disease genes.COPYRIGHT KIPO 2017본 발명은 Transactive response(TAR) DNA binding protein-43(TDP-43)-GAL4 초파리 돌연변이체와 TDP-43-GAL4 및 UAS-Green fluorescent protein(GFP) 초파리 이중돌연변이체 및 이의 제조방법에 관한 것으로서, 더욱 상세하게는 형광단백질을 초파리 TDP-43 유전자가 발현되는 조직에서 발현시킬 수 있으며, 이를 이용해 초파리 TDP-43 유전자의 발현 위치 및 발현양상을 확인할 수 있는 TDP-43-GAL4 및 Green fluorescent protein (GFP) 과발현 이중돌연변이 초파리 모델 제작 및 이의 제조방법을 포함한다.상기와 같은 본 발명에 따르면, 유전자 전사를 특정 조직에서 과발현시킬 수 있는 발현시스템인 GAL4 시스템을 이용함으로써 초파리 게놈으로부터 TDP-43 유전자의 프로모터 지역을 분리하여 제작한 TDP-43 유전자의 조직특이적 발현 돌연변이 및 형광단백질 과발현 돌연변이를 동시에 포함하는 초파리 이중돌연변이체를 및 이의 제조방법을 제공하며, 상기 초파리 이중돌연변이체는 TDP-43 관련 인자의 스크리닝 뿐만 아니라 초파리의 TDP-43 유전자의 기능 및 사람의 퇴행성 뇌 질환 유전자 기능에 관한 연구에 매우 유용한 동물 모델로 이용될 수 있다.
