The present invention relates to a recombinant adenovirus vector that produces immunity against an enterovirus infection. In one embodiment, a recombinant adenoviral vector of the invention comprises an expression cassette encoding the P1 protein of the enterovirus and the 3CD protease. In another embodiment, a recombinant adenoviral vector of the invention comprises an expression cassette encoding the 3C protease or 3CD protease of an enterovirus. The present invention also relates to a vaccine composition comprising a recombinant adenovirus vector as described. There is provided a method of inducing an immune response in a subject against an enterovirus infection using a recombinant adenoviral vector and a vaccine composition.
