Use of oleuropein (I) or its derivatives to prepare a composition for stimulating formation of bone and/or for inhibiting resorption of bone, in humans or animals. An independent claim is also included for a nutritional or pharmaceutical (human or veterinary) composition for stimulating formation of bone and/or for inhibiting resorption of bone, in which the active ingredient is (I) or one of its derivatives. ACTIVITY : Osteopathic； Antiarthritic； Cytostatic. Age-related osteoporosis (with inflammation) was induced in 6 month old rats by removal of their ovaries and after 7 days they were fed (a) base ratio (containing 2.5% each of groundnut and rapeseed oils)； (b) the base ration plus 0.5% (I) or (c) the base ration plus 5% extra virgin olive oil, for 72 days. On day 55, inflammation was induced by subcutaneous injection of talc suspended in physiological saline, and on day 80 the animals were killed and examined. The mean mineral density of bone (g/m 2>；) was 0.211 in (a)； 0.226 in (b) and 0.225 in (c)； compare 0.238 in sham-operated animals (X) given the base ration. Concentrations of the inflammatory marker orosomucoid (Microg/l) were 35.9 in (a), 17.8 in (X) and 22.8 in (c), and treatment with (I) also reduced splenomegaly. MECHANISM OF ACTION : (I) is already known to have antioxidant, antiinflammatory, antifungal, antibacterial and antiviral activities, but has now been found to increase density of both trabecular and cortical bone, by stimulating both formation of the protein network and mineralization.
