Methods of treating a patient having or at risk of developing an autoimmune disease (e.g., type 1 diabetes) are provided. The method comprise administering to the patient a composition containing a CD21 agonist. The CD21 agonist may be a polypeptide, an antibody or an antigen-binding fragment thereof, a nucleic acid molecule, or a small molecule. The CD21 agonist induces expression of TNF-a and/or intracellular signaling through NF-KB upon binding to CD21 receptor in a cell of the patient.
