A method of generating an isolated population of cells comprising anti-third party cells having a central memory T-lymphocyte (Tcm) phenotype, the cells being tolerance-inducing cells and/or endowed with anti-disease activity, and capable of homing to the lymph nodes following transplantation is disclosed. The method comprising: (a) contacting peripheral blood mononuclear cells (PBMC) with a third party antigen or antigens in the presence of IL-21 so as to allow enrichment of antigen reactive cells； and (b) culturing the cells resulting from step (a) in the presence of IL-21, IL-15 and IL-7 in an antigen free environment so as to allow proliferation of cells comprising the central memory T-lymphocyte (Tcm) phenotype.중심 기억 T-림프구(Tcm) 표현형을 가지는 항-제3자 세포를 포함하는 세포군 분리물의 제조 방법으로서, 상기 세포는 관용성-유도성 세포 및/또는 항-질환 활성이 부여된 세포로서 이식 후에 림프절로 회귀할 수 있는, 방법을 개시한다. 상기 방법은: (a) 말초 혈액 단핵 세포(PBMC)를 IL-21의 존재 하에 제3자의 항원 또는 항원들과 접촉시켜, 항원 반응성 세포를 농화하는 단계； 및 (b) 상기 (a) 단계로부터 생성되는 세포를 무-항원 환경 하에 IL-21, IL-15 및 IL-7의 존재 하에 배양하여, 중심 기억 T-림프구(Tcm) 표현형을 가지는 세포를 증식시키는 단계를 포함한다.
