The present invention relates to tumor necrosis factor-alpha (TNF-alpha)/C-X-C motif chemokine 10 (CXCL-10) double targeting antibody based on the IgG format. Specifically, it was verified that an antibody, in which scFv having a heavy chain variable region and a light chain variable region of the CXCL10-specific antibody binds to the C-terminus of the heavy chain constant region of the TNF-alpha-specific antibody, is a bispecific antibody which effectively binds to both TNF-alpha and CXCL10, thus the antibody can be useful as a double targeting antibody capable of identifying TNF-alpha/CXCL10. A composition of the present invention comprises a TNF-alpha/CXCL-10 double targeting antibody which effectively binds to both TNF-alpha and CXCL10. The double targeting antibody of the present invention has excellent TNF-alpha inhibitory activity and osteoclast differentiation inhibitory activity when compared with the TNF-alpha or CXCL10 single targeting antibody. The composition of the present invention can be used in prevention or treatment of immune disease.본 발명은 IgG 포맷 기반의 TNF-α/CXCL-10 이중 타겟 항체에 관한 것으로 구체적으로 CXCL10 특이적 항체의 중쇄 가변영역 및 경쇄 가변영역을 갖는 scFv를 TNF-α 특이적 항체의 중쇄 불변영역 C-말단에 결합된 형태의 항체는 TNF-α 및 CXCL10에 모두 효과적으로 결합하는 이중 특이성 항체임을 확인함으로써, 상기 항체는 TNF-α/CXCL10을 인식할 수 있는 이중 타겟 항체로서 유용하게 사용될 수 있다. 본 발명의 조성물은 TNF-α(tumor necrosis factor-alpha) 및 CXCL10(C-X-C motif chemokine 10)에 모두 효과적으로 결합하는 TNF-α/CXCL-10 이중 타겟 항체를 포함한다. 본 발명의 이중 타겟 항체는 TNF-α 또는 CXCL10 단일 타겟 항체와 비교하여 우수한 TNF-α 억제능 및 파골세포 분화 억제능을 갖는다. 본 발명의 조성물은 면역질환의 예방 또는 치료에 이용될 수 있다.
