A humanized monoclonal antibody (mAb) or fragment thereof that specifically binds to a Tomsen-Friedenreich (TF) human tumor antigen is provided. Three different variable heavy chains and three variable light chains are provided, which can be combined, making a total of 25 different heavy and light chain combinations. Methods for using the mAb and fragments thereof for cancer treatment and imaging are provided, as are methods for making the mAb or fragments thereof. In vitro cell cultures and kits expressing the mAbs and fragments thereof are also provided.
