Provided herein are methods of activating immune cells in a subject. In some embodiments, the method comprises passing immune cells from the subject through an immune regulatory chamber comprising tumor cells to activate the immune cells and return the activated immune cells to the subject. In some embodiments, the method further comprises isolating the immune cell-containing portion of the sample and passing the immune cell-containing portion through the immune regulatory chamber.
