Methods are provided for producing picornavirus-like particles (PVLPs) in plants. The method includes introducing a first nucleic acid and a second nucleic acid into a plant, plant part, or plant cell. The first nucleic acid includes a first regulatory region active in a plant operably linked to a nucleotide sequence encoding a picornavirus polypeptide. The second nucleic acid is active in the plant and includes a second regulatory region operably linked to a nucleotide sequence encoding one or more picornavirus proteases. Plants, plant parts, or plant cells are incubated under conditions that allow expression of the nucleic acid, thereby producing PVLP.
