PROBLEM TO BE SOLVED: To provide compositions for treating HER2-expressing cancer in patients.SOLUTION: Provided herein is a composition comprising a main species HER2 antibody that binds to domain II of HER2 and an amino acid sequence variant thereof comprising an amino-terminal leader extension. Further provided is the composition where the amino-terminal leader extension is present in the light chain of the antibody variant, and the main species HER2 antibody and the antibody variant are both intact antibodies, as well as a pharmaceutical formulation comprising the composition.
