The present disclosure relates to the antibody engineering field and, moreparticularly, to a process for thescreen-ing of antibodies and/or the modulation of the agonistic/antagonisticactivity of antibodies. More particularly, the disclosurecon-cerns a process of improving the antagonistic activity of a monoclonalantibody directed against a specific target molecule, or adivalent functional fragment or derivative thereof, said antibody beingcapable of inhibiting one or more of the biological activitiesof said target molecule, wherein said process comprises a stage ofreconfiguration of the hinge region consisting of a modificationof the amino acid sequence of said hinge region by the deletion, the additionor the substitution of at least one amino acid. Thedisclosure also relates to polypeptides useful for such a modulation methodand the obtained antibodies.
