The present invention relates to a composition for preventing or treating cancer, containing leucine zipper/EF hand-containing transmembrane protein 1 (LETM1) or a gene encoding the same. The composition containing LETM1 according to the present invention can induce the selective breakage of cancer cell mitochondria caused by ATP deficiency in addition to the activation of AMP-activated protein kinase (AMPK), can change an Akt signal, can suppress cell cycle and can promote apoptosis.
