The method amplified a PEDV S2A gene fragment, and then prokaryotically expressed and purified to obtain the recombinant S2 protein. Based on this, the inventors also established a method for preparing a recombinant S2 protein polyclonal antibody, that is, to immunize animals to obtain a polyclonal antibody serum by emulsifying the recombinant S2 protein with Freund's adjuvant. Experiments results showed that this invention successfully amplified part region with strong antigenicity of S2 gene of PEDV epidemic mutant strain CH/GX/2015/750A, and obtained recombinant S2 protein by prokaryotical expression.. In addition, the reactivity and immunogenicity of the recombinant S2 protein were preliminarily studied, which laid a foundation for the further study of the function of PEDV S2 protein and the development of PEDV diagnostic kit or PEDV subunit vaccine. 1
