Described herein is a method of evaluating a subject for the presence of orsusceptibility toa cancer associated with an IDH1-G97D mutant enzyme with 2-hydroxyglutarate(2HG)neoactivity. The method comprises: a) analyzing the subject, or a tissue,product or bodilyfluid sample from the subject, for the presence of 2HG by a chromatographicmethod； andb) analyzing for the presence in the subject or sample from the subject of anIDH1-G97Dmutant protein or a DNA or RNA encoding an IDH1-G97D mutant enzyme which has2HG neoactivity by DNA sequencing, immunoanalysis or enzymatic activityassays；where the presence of 2HG is indicative that the subject has or is susceptibleto a cancerassociated with an IDH1-G97D mutant enzyme with 2HG neoactivity.
