Provided are compositions and methods for isolating L-glutosinate from compositions comprising L-glutosinate and glutamate. The method comprises converting glutamate to pyroglutamate, then isolating L-glutosinate from pyroglutamate and other components of the composition to obtain substantially purified L-glutosinate. A composition comprising L-glufosinate and glutamate is subjected to elevated temperature for a sufficient period of time to allow glutamate to be converted to pyroglutamate, followed by isolation of L-glutosinate from pyroglutamate and other components of the composition to substantially purified L -Glufosinate is obtained. Alternatively, glutamate can be converted to pyroglutamate by enzymatic conversion. The purified L-glutosinate is present in the final composition at a concentration of at least 90% of the sum of L-glutosinate, glutamate and pyroglutamate. In some embodiments, some of the glutamate in the starting composition can be separated from L-glutosinate using a crystallization step.
