The present invention relates to one-time spore forming strains and a preparing method thereof. The one-time spore forming strains can lose the ability to form spores after forming spores once by removing the spo0A genes being a major regulator of spore formation in a spore formation process. More specifically, a recognition sequence of the site-specific recombinase is inserted into the upstream and downstream of the spo0A gene present in the chromosome of strains of Bacillus species forming the spores. A promoter specifically expressed only in forespores in the sporozoite period and the open-reading frame (ORF) of the site-specific recombinase being expressed as the promoter are inserted into the chromosome.본 발명은 포자 형성 과정 중 포자 형성 시작의 주요 조절인자인 spo0A 유전자가 제거되어 한 번 포자를 형성하면 포자를 형성하는 능력을 상실하는 단회 포자 형성 균주 및 이의 제조방법에 관한 것으로, 더욱 상세하게는 포자를 형성하는 바실러스 속 균주의 염색체 상에 존재하는 spo0A 유전자의 업스트림(upstream) 및 다운스트림(downstream)에 부위 특이적 재조합효소(site-specific recombinase)의 인식 서열이 삽입되고, 포자 형성기에 전포자(forespore)에서만 특이적으로 발현되는 프로모터와 상기의 프로모터로 발현되는 부위 특이적 재조합효소(site-specific recombinase)의 ORF(open-reading frame)가 염색체에 삽입된 것을 특징으로 하는 단회 포자 형성 균주 및 이의 제조방법에 관한 것이다.
