The present invention relates to a method of producing rough strains of abacterium, such as Mycobacterium obuense, said method comprising exposing saidbacterium to a sulfone and/or sulfonamide (such as 4,4'-Diaminodiphenylsulfoneor an analogue thereof). A rough strain of Mycobacterium obuense producible bysaid method and uses thereof. In particular, uses of a rough strain ofMycobacterium obuense deposited under the Budapest Treaty at the NCTC withthe accession number NCTC 13365 to modulate an immune response are alsodisclosed.
