A separation method for A2-β-casein in milk. Considering the characteristics of the beta-casein comprehensively, such as isoelectric point, Ca2+ ion sensitivity and low-temperature low-acid free state, a purification method for the A2-β-casein is explored, and the A2-β-casein having a purity of 85%t or higher may be obtained by means of fast separation and enrichment. The method may be used for separation and purification of the A2-β-casein, and may also be used for the separation and purification of A1-β-casein or a mixed β-casein comprising both the A1-β-casein and the A2-β-casein. The problems of long beta-casein separation time, complicated steps and high cost in the existing technology are solved, and the conditions are provided for detecting the β-casein types in milk products. In addition, the process is simple, the cost is low, and the method is thus applicable to large-scale production.L'invention concerne un procédé de séparation de l'A2-ß-caséine dans le lait. Compte tenu des caractéristiques de la bêta-caséine dans sa globalité, comme le point isoélectrique, la sensibilité aux ions Ca2+ et l'état libre faiblement acide à basse température, un procédé de purification pour l'A2-ß-caséine est exploré, et l'A2-ß-caséine ayant une pureté égale ou supérieure à 85 %t peut être obtenue au moyen d'une séparation et d'un enrichissement rapides. Le procédé peut être utilisé pour la séparation et la purification de l'A2-ß-caséine et peut également être utilisée pour la séparation et la purification de l'A1-ß-caséine ou d'une ß-caséine mélangée comprenant à la fois l'A1-ß-caséine et l'A2-ß-caséine. Les problèmes de temps de séparation long de la ß-caséine, d'étapes compliquées et du coût élevé dans la technologie existante sont résolus, et les conditions sont réunies pour détecter les types de ß-caséine dans des produits laitiers. De plus, le procédé est simple, le coût est faible et le procédé est ainsi applicable à la production à grande échelle.
