The invention provides an ex-vivo method, i.e. an in vitro method, for genetically modifying cells of a vascularised tissue, which can be an organ, during normothermic or subnormothermic perfusion without damaging the tissue or its cells due to oxygen undersupply. The method enables keeping the vascularised tissue at normothermic or subnormothermic conditions over a sufficient period of time to genetically modify the cells e.g. with viral vectors.L'invention concerne un procédé ex vivo, c'est-à-dire un procédé in vitro, pour modifier génétiquement des cellules d'un tissu vascularisé, qui peut être un organe, pendant une perfusion normothermique ou subnormothermique sans endommager le tissu ou ses cellules du fait d'un apport insuffisant d'oxygène. Le procédé permet de garder le tissu vascularisé dans des conditions normothermiques ou subnormothermiques pendant une période de temps suffisante pour modifier génétiquement les cellules, par exemple, avec des vecteurs viraux.
