The present invention relates to a composition for anti-cancer vaccine, which is prepared by homogenizing artificial antigen expression cells expressing different immunization molecules commencing MHC using insect cells, isolating and purifying cell membrane-derived endoplasmic reticulum, treating the obtained endoplasmic reticulum with paraformaldehyde (PFA), and fixing peptide specific to MHC and T cell receptor (TCR) expressed in the endoplasmic reticulum. According to the fixation, an effect of semipermanently stabilizing MHC/peptide complex having low binding strength is obtained, so that an effect of maintaining immunizing function of endoplasmic reticulum for a long time is obtained.본 발명의 곤충세포를 이용하여 MHC를 위시한 서로 다른 면역 분자들을 발현하는 인공항원발현세포를 homogenize하고, 이때 만들어지는 세포막 유래 소포체를 분리 정제한 후, 얻어진 소포체를 파라포름알데하이드(PFA)로 처리하여 소포체에서 발현되는 MHC와 T세포 수용체(T Cell Receptor, TCR) 특이적 펩타이드를 고정화시킨항암 백신용 조성물에 관한 것이다. 상기 고정화에 의해서, 결합강도가 낮은 MHC/peptide complex를 반영구적으로 안정화 시키는 효과가 있으며, 이로 인해 소포체의 면역기능이 오래 지속될 수 있는 효과를 가져올 수 있다.
