Disclosed is in vitro cell culture comprising human cells wherein at least 5% of said human cells are endocrine precursor cells that express a marker selected from the group consisting of neurogenin 3 (NEUROG3), paired box 4 (PAX4) and NKX2 transcription factor related locus 2 (NKX2.2), said endocrine precursor cells being multipotent cells that can differentiate into pancreatic islet hormone-expressing cells that express at least one pancreatic hormone selected from the group consisting of insulin, somatostatin, ghrelin, pancreatic polypeptide and glucagon. Also disclosed is an in vitro method of producing endocrine precursor cells, said method comprising the steps of: (a) providing a cell population comprising pluripotent human cells with a TGF-beta superfamily growth factor, thereby producing definitive endoderm cells; (b) withdrawing TGF beta superfamily from said cell population; (c) providing said cell population with a retinoid, thereby producing human PDX1-positive pancreatic endoderm cells; and (d) incubating said cell population in a culture medium for sufficient time to permit formation of human endocrine precursor cells expressing neurogenin 3 (NEUROG3).
