Human biliary tree stem / progenitor cells (hbtscs) are being used for cell therapies in patients with liver cirrhosis. a cryopreservation method has been established to optimize the supply of hbtscs for these clinical programs and which comprises serum-free kubota medium (km) supplemented with 10% dimethyl sulfoxide (dmso), recombinant human albumin -3% and hyaluronans 0.1 %. cryopreserved hbtscs versus newly isolated ones were similar in vitro with respect to self-replication, traits of severity and multipotency. they were able to differentiate into hepatocytes, cholangiocytes or functional pancreatic islets, generating similar levels of albumin secretion or glucose-inducible insulin levels. cryopreserved versus newly isolated hbtscs were also capable of grafting into immunocompromised mice generating cells with specific gene expression for humans and levels of human albumin in murine serum that were higher for cryopreserved hbtscs than for newly isolated hbtscs. the successful cryopreservation of hbtscs facilitates the establishment of hbtscs cell banks that offer logistical advantages for clinical programs for the treatment of liver disease.células-tronco/progenitoras da árvore biliar humana (hbtscs) estão sendo usadas para terapias celulares de pacientes com cirrose hepática. foi estabelecido um método de criopreservação para otimizar o abastecimento de hbtscs para esses programas clínicos e que compreende meio de kubota livre de soro (km) suplementado com sulfóxido de dimetila 10% (dmso), albumina humana recombinante -3% e hialuronanos 0,1%. hbtscs criopreservadas versus recém isoladas foram similares in vitro com relação à auto-replicação, traços de severidade e multipotência. elas foram capazes de se diferenciar em hepatócitos, colangiócitos ou ilhotas pancreáticas funcionais, gerando níveis similares de secreção de albumina ou de níveis de insulina induzíveis por glicose. hbtscs criopreservadas versus recém isoladas foram igualmente capazes de enxertia em c
