A method of treating a disease in a subject in need thereof is disclosed. The method comprising: (a) transplanting a non-syngeneic cell or tissue graft to the subject and (b) administering to the subject a therapeutically effective amount of an isolated population of cells comprising non-graft versus host (GVHD) inducing anti-third party cells having a central memory T-lymphocyte (Tcm) phenotype, the cells being tolerance-inducing cells and capable of homing to the lymph nodes following transplantation, and further wherein the cells are either: (i) non-syngeneic with both the subject and the graft or (ii) non-syngeneic with the graft and syngeneic with the subject, thereby treating the subject.Linvention concerne une méthode de traitement dune maladie chez un sujet dont létat nécessite un tel traitement. La méthode consiste à: (a) transplanter chez le sujet un greffon non syngénique cellulaire ou tissulaire et (b) administrer au sujet une quantité thérapeutiquement efficace dune population isolée de cellules comprenant des cellules anti-tiers et ninduisant pas de réaction du greffon contre lhôte, lesdites cellules possédant un phénotype de lymphocyte T (Tcm) de mémoire centrale, ces cellules étant des cellules induisant une tolérance et pouvant migrer vers les ganglions lymphatiques après transplantation. En outre, les cellules sont: (i) soit des cellules ne présentant pas de syngénicité avec le sujet et le greffon (ii) soit des cellules ne présentant pas de syngénicité avec le greffon, mais en présentant avec le sujet, permettant ainsi de traiter ce dernier.
