Disclosed are a polyconjugate and a preparation method thereof, wherein the polyconjugate comprises an active drug and a macromolecule polymer, with the active drug and the macromolecule polymer being linked via a chemical bond, wherein the active drug is a polyene macrolide antibiotic and the macromolecule polymer is selected from a polyamino acid and a polyamino hexose. The polyconjugate displays good properties in aspects of biology and physical and chemical properties, for example, the polyconjugate can improve the solubility of an active drug.
