The present invention provides recombinant replication-defective adenoviralvectors derived from chimpanzee adenoviruses and methods for generatingrecombinant adenoviruses in human E1-expressing cell lines. The invention alsoprovides compositions and methods suitable for use for the delivery andexpression of transgenes encoding immunogens against which a boosted immuneresponse is desired. The invention further provides methods of generatingclinical grade vector stocks suitable for use in humans. In a particularembodiment the invention contemplates the use of vectors comprising transgeneswhich encode tumor associated antigens in vaccines and pharmaceuticalcompositions for the prevention and treatment of cancer.
