The present invention relates to a composition for treating and preventing osteoporosis which includes phloretin which suppresses bone resorption of the osteoclast which promotes bone resorption due to osteoporosis. More specifically, the present invention relates to the composition for treating and preventing osteoporosis in which phloretin reduces the expression of carbonic anhydrase and V-ATPase D2 which participate in acidification condition during bone resorption of the osteoclast and suppresses secretion of MMP-9 secreted during bone resorption and suppresses bone resorption of the osteoclast by reducing the expression and secretion of Cathepsin K. In addition, bone density, which is reduced by ovariectomization in an ovariectomized animal model, is increased. Therefore, phloretin suppresses the differentiation of the osteoclast without side effects and suppresses bone resorption.COPYRIGHT KIPO 2014본 발명은 골다공증으로 인한 골흡수을 도모하는 파골세포의 골흡수를 억제시키는 phloretin을 함유하는 골다공증 치료 및 예방용 조성물에 대한 것이다. 보다 구체적으로, phloretin이 파골세포의 골흡수시 산성화 조건에 관여하는 Carbonic anhydrase 와 V-ATPase D2의 발현을 감소시키며 골흡수시 분비되는 MMP-9의 분비를 억제시키고, Cathepsin K 의 발현 및 분비를 감소시켜 파골세포의 골흡수를 억제시킨다. 또한 난소절제 골다공증 동물모델에서 난소절제로 인한 감소된 골밀도를 상승시켜주었다. 이로서 phloretin은 부작용이 없으면서 파골세포의 분화를 억제하여 골흡수를 억제시킬 수 있는 골다공증 치료 및 예방할 수 있는 조성물에 대한 것이다.
