The present invention relates to a method for proliferation of regulatory T cells (Treg) in vitro by using an autologous antigen-presenting cell (APC) and uses thereof. More specifically, the method, which is used for the in-vitro proliferation of the CD4+Treg, includes the steps of (a) preparing the CD4+Treg and CD4-PBMC respectively and (b) stimulating the CD4+Treg with the CD4-PBMC, anti-CD3 antibody, anti-CD28 antibody, interleukin2 (IL2), and TGF-b(beta) for proliferating the CD4+Treg. The present invention also relates to a composition for inhibiting the transplant rejection reaction and a pharmaceutical composition for the prevention or treatment of autoimmune diseases or inflammatory diseases.
