The present invention relates to a composition capable of effectively preventing, ameliorating and treating enlargement of the prostate gland. The vanillic acid of the present invention significantly reduced prostate tissue size and weight in an experimental animal model of benign prostatic hyperplasia and showed histological improvement effect including epithelial cell thickness. It also effectively reduced the expression of proteins or factors associated with the induction of prostatic hyperplasia.
