The present invention relates to novel siRNA, and a highly efficient double-stranded oligo RNA structure and nanoparticles containing the same. The double-stranded oligo RNA structure has a structure in which a hydrophilic material and a hydrophobic material are conjugated to both terminals of the double-stranded RNA (siRNA) by a simple covalent bond or a linker-mediated covalent bond to efficiently deliver the double-stranded RNA structure into cells, and may be changed into a nanoparticle form in an aqueous solution by hydrophobic interactions between the double-stranded RNA structures. The siRNA included in the double-stranded oligo RNA structure is preferably siRNA specific to fibrosis or a respiratory disease-associated gene, particularly, amphiregulin or stratifin. In addition, the present invention relates to a pharmaceutical composition including siRNA, and a highly efficient double-stranded oligo RNA structure and nanoparticles containing the siRNA, as an active ingredient for preventing or treating fibrosis or respiratory diseases. In addition, the present invention relates to a method for preventing or treating fibrosis or respiratory diseases including a step of administering the pharmaceutical composition for preventing or treating fibrosis or respiratory diseases to a subject in need thereof.본 발명은 신규한 siRNA, 이를 포함하는 고효율 이중나선 올리고 RNA 구조체 및 나노입자에 관한 것으로, 상기 이중나선 올리고 RNA 구조체는 세포 내로 효율적으로 전달되도록 하기 위하여, 이중나선 RNA(siRNA)의 양 말단에 친수성 물질 및 소수성 물질을 단순 공유결합 또는 링커-매개(linker-mediated) 공유결합을 이용하여 접합된 형태의 구조를 가지며, 수용액에서 상기 이중나선 올리고 RNA 구조체들의 소수성 상호작용에 의해 나노입자 형태로 전환될 수 있다. 상기 이중나선 올리고 RNA 구조체에 포함된 siRNA는 섬유증 또는 호흡기 질환 연관 유전자, 특히 엠피레귤린 또는 스트라티핀 특이적 siRNA인 것이 바람직하다. 또한, 본 발명은 siRNA, 이를 포함하는 고효율 이중나선 올리고 RNA 구조체 및 나노입자를 유효성분으로 포함하는 섬유증 또는 호흡기 질환의 예방 또는 치료용 약학조성물에 관한 것이다.또한, 본 발명은 섬유증 또는 호흡기 질환의 예방 또는 치료용 약학조성물을 섬유증 또는 호흡기 질환의 예방 또는 치료를 필요로 하는 개체에 투여하는 단계를 포함하는 섬유증 또는 호흡기 질환의 예방 또는 치료방법에 관한 것이다.
