The present invention relates to a recombinant haemagglutinin domain-containing protein having SEQ ID No.: 1 for the detection and diagnosis of glanders disease caused by Burkholderia mallei in equines. This protein is encoded by polynucleotide sequence having SEQ ID No.: 2 encoding said protein. The invention further relates to a process for preparing and purifying the recombinant haemagglutinin domain-containing protein comprising identifying the sequence coding for said protein amplifying the sequence of using genomic DNA of B. mallei digesting the amplified product with restriction enzymes cloning the digested product in prokaryotic expression system transforming the cloned product in an expression host and purifying the protein by immobilized metal affinity chromatography.
