Disclosed are a colorectal cancer solid tumour primary cell and colorectal cancer ascitic fluid primary tumour cell culturing method, and a matching reagent. The core of the present invention is: (1) using a mild cell dissociation reagent to treat colorectal cancer solid tumour tissue, and using a mild method to separate colorectal cancer cells in ascitic fluid, to ensure as far as possible the viability of the cancer cells； (2) preparing a special serum-free culture medium, and using a suspension culture system to perform in vitro culturing of tumour cells from a colorectal cancer solid tumour, to ensure cancer cell normal expansion while minimizing interference from normal cells.
