Sol-gel phase-reversible hydrogel templates are used to prepare discrete microstructures of predetermined size and shape. An aqueous solution of hydrogel-forming material is covered over the microfabricated silicon wafer master template with a predetermined microstructure, such as a filler. Hydrogel templates are generally prepared by lowering the temperature and stripping the prepared hydrogel template from the silicone master template. Wells of a predetermined size and shape on the hydrogel template are filled with a solution or paste of water-insoluble polymer and the solvent is removed to form a solid structure. The prepared microstructures simply dissolve the hydrogel template in water and release it from the hydrogel template. Microstructures are obtained by centrifugation. The microstructures produced in this way exhibit a predetermined size and shape that exactly corresponds to the microwells of the hydrogel template.
