The present invention provides an antibody and an antigen-binding fragment thereof that specifically bind to a polypeptide or an antigenic portion thereof, wherein the polypeptide is a) MUC16 ectodomain polypeptide, b) MUC16 cytoplasmic domain poly. Antibodies and antigen-binding fragments thereof selected from peptides and c) MUC16 extracellular domain polypeptides containing cysteine loop polypeptides. The antibodies of the present invention and compositions containing them are useful for diagnostic and therapeutic applications for diseases in which MUC16 is overexpressed, such as cancer.
