The present invention relates to a composition for assessment of Human Sperm Funtion using nuclear chromatin Decondensation (NCD) test, said composition comprising sodium chloride of concentration ranging between 0.05 to 0.4% w/v, Sodium dodecyle Sulfate (SDS) of concentration ranging between 0.5 to 2.0 % w/v, and Dithiotheritol (DTT) of concentration ranging between 0.03 to 0.06 % w/v a process for preparing the same and method thereof.
