PURPOSE: A method of culturing bacillus subtilis ch for protecting rust of plant roots is provided to reduce manufacturing cost, thereby mass-producing bacillus subtilis ch strain with low cost. CONSTITUTION: A method of culturing bacillus subtilis ch for protecting rust of plant roots comprises the following steps: collecting soil and spreading on agar medium； separating milky colored colonies from the agar medium； shake culturing each strain in Luria Bertani(LB) liquid culture medium； centrifuging and collecting the strain； suspending in sterile water of equivalent amount as the strain； and selecting strains which obstruct rearing of pathogen by confronting and cultivating with the bacteria which caused rust of plant roots.본 발명은 식용뿌리작물의 뿌리썩음병 방제용 바실러스 서브틸루스 시에치 균주의 배양 방법에 관한 것으로, 보다 상세하게는 식용뿌리작물에 뿌리썩음병을 일으키는 질병에 대한 방제를 위하여 국내 근권 토양에서 분리된 Bacillus subtillus CH균주를 산업적으로 대량 생산하기 위하여 고가의 배양 배지 대신 저렴한 배양용 배지를 이용하여 생산단가를 낮추고 효과를 배가 시킬 수 있는 식용뿌리작물의 뿌리썩음병 방제용 바실러스 서브틸루스 시에치 균주의 배양 방법에 관한 것이다.본 발명에 따른 식용뿌리작물의 뿌리썩음병 방제용 바실러스 서브틸루스 시에치 균주의 배양 방법은, 토양을 채취하여 한천 배지에 도말하는 단계； 한천 배지상에서 우윳빛의 단 콜로니를 분리하는 단계； 각 균주를 Luria Bertani(LB) 액체배지에서 진탕배양한 후 원심분리시켜 균을 회수하여 동량의 멸균수에 현탁하는 단계； 및 뿌리섞음병을 유발하는 윈인균과 대치배양하여 병원균의 생육을 저해하는 균주를 선발하는 단계를 포함하여 구성된 것을 특징으로 한다.
