In one aspect, provided herein is a composition comprising a polyorthoester, an organic acid, and a therapeutically active agent, e.g., one containing one or more amino groups, dispersed or solubilized in the composition. In some embodiments, the active agent is an anesthetic. In some further embodiments, the anesthetic is a local anesthetic.
