The present invention provides a method for producing a protein conjugate from a protein having at least two cysteines. In one embodiment, the protein with the disulfide linkage is reduced to provide two free cystines for reacting with 1,3-dihaloacetone or similar reactants that link the sulfur atoms of the two cystines together. Subsequently, a ketone inserted between the sulfur atoms is used to form a Schiff base for the aminated charge molecule, thereby conjugating the protein to the payload. In another embodiment, the two cysteine residues are tied together by reaction with 1,3-dihaloacetone or similar reactants.
