A biological activity composition containing Cortex Magnolia Officinalis hot-water extract stimulating CB2 expression in stomach tissue in the prevention or improvement of gastric ulcer is provided. The Magnolia bark raw material is added into a sterile water and heated to 100℃ for at least 20 minutes, and then subjected to filtration, concentration and lyophilization, thereby forming the Magnolia bark hot-water extract. The Magnolia bark hot-water extract in vitro specifically promotes the cell viability and decreases the production of reactive oxygen species (ROS) of the gastric epithelial cells in the acidic medium. In vivo, the Magnolia bark hot-water extract stimulates the expression of the cannabinoid receptor type 2 (CB2) and decreases the production of tumor necrosis factor-α (TNF-α) and interleukin 6 (IL-6) of gastric tissue and thus, significantly decreases the ulcer index and elevates the ulcer protection to prevent or improve gastric ulcer.本發明揭露一種厚朴熱水萃取物用於製備防治或改善胃潰瘍之生物活性組成物。係將厚朴原料與無菌水加熱至100℃且持溫至少20分鐘,再依序進行過濾步驟、濃縮步驟與冷凍乾燥步驟,以形成厚朴熱水萃取物。厚朴熱水萃取物於活體外於酸性環境中促進胃上皮細胞株之細胞存活率及降低自由基之生物活性,以及於活體內提昇胃組織CB2受體基因表現量,且減少胃組織TNF-α以及IL-6之含量,可顯著降低胃潰瘍指數、提升胃保護效果,以防治或改善胃潰瘍。
