Offenbart wird eine optimierte pharmazeutische Formulierung für die Behandlung von entzündlichen Erkrankungen des Kolons, wobei die pharmazeutische Formulierung eine Pellets enthaltende Kapsel ist, die zur oralen Anwendung geeignet ist und den Wirkstoff gezielt an den Wirkort Kolon bringt. Erreicht wird dies durch eine komplexe und multiple Umhüllung von Pellets, die eine modifizierte Wirkstofffreisetzung ermöglichen. Erst im Kolon ist die Freisetzung des Wirkstoffes maximal, bei gleichzeitig niedrigen Blutplasmaspiegeln. Die Ergebnisse der pharmazeutischen Untersuchungen zur in-vitro Freisetzung werden durch die Ergebnisse in pharmakokinetischen sowie klinischen Studien und der dort gezeigten klinischen Wirksamkeit bestätigt.
