The present invention relates to a pharmaceutical composition for enhancing sensitivity to anticancer drug, comprising an inhibitor for activity or expression of kallikrein 6 (KLK6) as an active ingredient. The present invention further relates to a method for screening anticancer drug sensitivity enhancer or anticancer drug resistance inhibitor against cancer which is tolerant to anticancer drug. More specifically, it has been found that auranofin decreases survival rate of gastric carcinoma cells deficient in KLK6, and the expression of KLK6 due to activity of p53 increases induction of autophagy. In addition, gene blockage due to methylation on a promoter causes reduction in the number of autophagy and drug sensitivity as well. Therefore, the composition of the present invention can be used in the anticancer drug sensitivity enhancer and a screening method thereof via inhibition of autophagy activity using inhibitor for activity or expression of KLK6 and LC3B.본 발명은 KLK6(kallikrein 6)의 발현 또는 활성 억제제를 유효성분으로 함유하는 항암제 민감성 증진용 및 항암 보조용 약학적 조성물, 그리고 항암제 내성 암에 대한 항암제 내성 억제제 또는 항암제 민감성 증진제의 스크리닝 방법에 관한 것으로, 구체적으로 오라노핀(auranofin)이 KLK6가 결핍된 위암 세포의 생존율을 감소시키는 것을 확인하였으며, p53의 활성으로 인한 KLK6의 발현은 오토파지 유도를 증가시키고, 프로모터 메틸화에 의한 유전자 차단은 오토파지 저감과 함께 약물 민감도를 줄이는 것을 확인하였다. 따라서, 본 발명은 KLK6 및 LC3B의 발현 또는 활성 억제제를 이용한 오토파지 활성 억제를 통해 항암제 민감성 증진제 및 이의 스크리닝 방법에 사용될 수 있음을 확인하였다.
