The present invention relates to a composition for preventing and treating an ischemic heart disease having an adeno-associated virus (AAV) as an active component including myocardial conversion genes and a cardiovascular generative gene, and more specifically, to a composition which has Gata4, Mef2c, and Tbx5, which are the myocardial conversion genes, and has Thymosin &beta；4, which is the cardiovascular generative gene. According to the present invention, the composition uses the adeno-associated virus which is clinically appropriate, and applies the Gata4, Mef2c, and Tbx5 (GMT) and Thymosin beta4 (T&beta；4) which is revascularization, differentiation, and cardiovascular generative gene capable of directly converting a fibroblast into a myocardial cell as a functional gene delivered inside to an ischemic disease. Therefore, the present invention enables to directly convert a cell of a myocardial infarction part covered by the fibroblast and an extracellular matrix into the myocardial cell, and to induce a cardiovascular generation by stimulating a neighboring cell related to revascularization and myocardial generation.본 발명은 심근전환유전자 및 심혈관재생유전자를 포함하는 아데노부속바이러스를 유효성분으로 포함하는, 허혈성 심장질환 예방 또는 치료용 조성물에 관한 것이다. 보다 구체적으로, 본 발명은 심근전환유전자인 Gata4, Mef2c, Tbx5 및 심혈관재생유전자인 Thymosin β4가 삽입된 아데노부속바이러스를 유효성분으로 포함하는 허혈성 심장질환의 예방 또는 치료용 조성물에 관한 것이다. 본 발명에 따른 상기 조성물은 임상적으로 적합한 아데노부속바이러스를 이용하되 그 내부에 전달되는 기능유전자 후보로써 섬유아세포를 심근세포로 직접전환할 수 있는 직접전환유전자인 Gata4, Mef2c, Tbx5 (이하 GMT) 및 혈관재생 및 분화 및 심근재생촉진유전자인 Thymosin beta4 (이하 Tβ4)를 허혈성 질환에 적용함으로써, 섬유아세포 및 세포외기질에 싸여 있는 심근경색부위의 세포를 심근세포로 직접전환함과 동시에 주위의 혈관재생 및 심근재생관련 세포 등을 자극하여 심혈관재생을 유도할 수 있다.
